Use of CAR-transduced natural killer cells in CD19-positive lymphoid tumors
Background Anti-CD19 chimeric antigen receptor (CAR) T-cell therapy has shown remarkable clinical efficacy in B-cell cancers. However, CAR T cells can induce substantial toxic effects, and the manufacture of the cells is complex. Natural killer (NK) cells that have been modified to express an anti-CD19 CAR have the potential to overcome these limitations. Methods In this phase 1 and 2 trial, we administered HLA-mismatched anti-CD19 CAR-NK cells derived from cord blood to 11 patients with relapsed or refractory CD19-positive cancers (non-Hodgkin’s lymphoma or chronic lymphocytic leukemia [CLL]). NK cells were transduced with a retroviral vector expressing genes that encode anti-CD19 CAR, interleukin-15, and inducible caspase 9 as a safety switch. KTE-X19 CAR T-cell therapy in relapsed or refractory mantle-cell lymphoma
Background Patients with relapsed or refractory mantle-cell lymphoma who have disease progression during or after the receipt of Bruton’s tyrosine kinase (BTK) inhibitor therapy have a poor prognosis. KTE-X19, an anti-CD19 chimeric antigen receptor (CAR) T-cell therapy, may have benefit in patients with relapsed or refractory mantle-cell lymphoma. Methods In a multicenter, phase 2 trial, we evaluated KTE-X19 in patients with relapsed or refractory mantle-cell lymphoma. Patients had disease that had relapsed or was refractory after the receipt of up to five previous therapies; all patients had to have received BTK inhibitor therapy previously. Patients underwent leukapheresis and optional bridging therapy, followed by conditioning chemotherapy and a single infusion of KTE-X19 at a dose of 2×106 CAR T cells per kilogram of body weight. A measurement of the Higgs boson mass in the diphoton decay channel
A measurement of the mass of the Higgs boson in the diphoton decay channel is presented. This analysis is based on 35.9 fb− 1 of proton-proton collision data collected during the 2016 LHC running period, with the CMS detector at a centre-of-mass energy of 13 TeV. A refined detector calibration and new analysis techniques have been used to improve the precision of this measurement. The Higgs boson mass is measured to be m H= 125.78±0.26 GeV. This is combined with a measurement of m H already performed in the H→ ZZ→ 4 ℓ decay channel using the same data set, giving m H= 125.46±0.16 GeV. This result, when further combined with an earlier measurement of m H using data collected in 2011 and 2012 with the CMS detector, gives a value for the Higgs boson mass of m H= 125.38±0.14 GeV. This is currently the most precise measurement of the mass of the Higgs boson.
